No, I just compiled LibTIFF with 12-bit JPEG support, but had no sample pictures to try with.

It should be no problem to update the patch for arbitrary bpp values less or equal 16.

What about adding support for PAM format?

There is no real need for direct PAM format support, as pamtopnm exists and does not affect the picture quality. You need pamtopnm anyway to read the plain text variants of PBM, PGM and PPM.

For full support of the range of Netpbm file formats, we'd better link against libnetpbm. (This, however, would make the dependency between LibTIFF and Netpbm circular ;-) .)
