Molecular Dynamics corporation uses 16-bit TIFF for images of 
electrophoretic gels, obtained from their Phosphorimagers.  I think it's
TIFF -- at least they provide a utility to export 8-bit TIFF.  They at 
one time had a modified NIH Image which could handle 16-bit.  I don't 
know if they would be willing to help you.
