Does the problem have anything to do with the library being built as a 
universal binary?

I am using libtiff 3.8.0 on an Apple G5 without noticing any endian 
issues.   I am using libtiff 3.9.0beta on a Sun SPARC system without 
issues.  However, I do believe that 3.8.2 had severe issues on the Sun 
system.

It is not uncommon for various libtiff releases to experience issues 
on big-endian systems.
