I strongly suspect you mean channels, and not layers, as in R-G-B for example. Is that correct? If not, if you do mean true layers, then you're in for an adventure, as there is no standard way to specify layers. Instead, there are mutually incompatible proprietary vendor-specific ways. So if you do mean true layers, you need to specify which application created the TIFF file.
