Jim,

I would review the configure output carefully to try and establish if
everything necessary for large file access is being done.  In my output
on 64bit linux I see:

checking for special C compiler options needed for large files... no
checking for _FILE_OFFSET_BITS value needed for large files... no
...
   C compiler:                         gcc -g -DDEBUG -Wall -W
   C++ compiler:                       g++ -g -DDEBUG

which seems related.  Beyond this you could try and write a small test program
that uses the low level TIFF io layer to seek to 2.5GB and read something and
see if it gets the right result as a way of estalishing whether this is the
issue.
